Creative Biolabs is a trusted partner in half-life extension services. Our scientists have rich experience in this field, and we can provide multiple strategies of half-life extension to meet the unique requirements of every client. The short half-life of therapeutic molecules restricts their widespread applications. Half-life extension has been recognized as a promising strategy to broaden the uses of biotherapeutics by increasing the time between applications. Various half-life extension strategies have been utilized for generating longer therapeutics. Covalently binding therapeutic molecules to albumin is a potential method to improve their pharmacokinetic features.

Introduction of Covalent Attachment to Albumin

The covalent bond between the natural albumin and the therapeutic molecules can be formed in situ. The main method of in situ covalent attachment of albumin utilizes the 34 amino acid cysteine of albumin. Furthermore, covalently binding exogenous albumin with chemotherapeutics can obtain the same benefits as the binding of endogenous albumin.

Applications of Covalent Attachment

The covalent attachment has been used to improve the pharmacokinetic properties of a wide range of target molecules therapeutics ranging from small molecule drugs to biologics. Small molecules drugs such as methotrexate, doxorubicin, and curcumin are coupled to albumin in the development of anticancer and anti-rheumatic therapeutics. A variety of peptides including Exendin-4 and insulin have been successfully manufactured. In addition, peptides were modified to formate the conjugation via new technology.
